چکیده اصلی

The therapy to treat the Human Immunodeficiency virus (HIV) and decrease the viral load has been a challenge since its discovery due to its ability to mutate, escape from drug therapies, and immunologic system. Consequently, the development of new therapies, especially potential molecules that focus on specific viral mechanisms to block viral replication, became a critical challenge in recent decades. The Reverse Transcriptase (RT) is an RNA- and DNA-dependent DNA polymerase enzyme that is responsible for transcribing viral RNA into double-stranded DNA. Non-nucleoside RT Inhibitors perform important role in HIV therapy, and they are responsible for inhibiting RT by blocking its allosteric site, which shows physico-chemical properties such as high hydrophobicity and key amino acid-to-hydrogen bond interactions. For this reason, designing new molecules with innovative heterocycles compatible with these characteristics can provide new potential inhibitors. For example, pentacyclic heterocycles like thiazole and its derivatives like thiazolidinedione, which have physico-chemical properties that can interact with the allosteric site of RT, can be a new perspective in designing new Non-nucleoside RT Inhibitors. This work reviews the scientific literature concerning these compounds and can help the design of new antiviral therapeutics.

این مقاله به بررسی خواص شیمیایی تریازول و تریازولیدین به عنوان بلوک‌های سازنده برای طراحی مهارکننده‌های RT غیرنوکلئوزیدی (NNRTI) می‌پردازد. این ترکیبات به دلیل تعامل با سایت آلوستریک آنزیم RT، پتانسیل بالقوه برای درمان HIV را دارند. هدف این مطالعه ارائه یک مرور ادبیات برای کمک به طراحی داروهای جدید است.

درمان ویروس ایمونودفیسینس انسانی (HIV) و کاهش بار ویروسی از زمان کشف آن به دلیل توانایی جهش، فرار از داروها و سیستم ایمنی، چالشی بوده است. در نتیجه، توسعه درمان‌های جدید، به‌ویژه مولکول‌های بالقوه که بر مکانیسم‌های ویروسی خاص تمرکز دارند تا تکثیر ویروس را مسدود کنند، در دهه‌های اخیر به چالش حیاتی تبدیل شده است. آنزیم معکوس‌نویس (RT) یک پلیمراز دی‌ان‌ای وابسته به RNA و DNA است که مسئول ترجمه RNA ویروس به دی‌ان‌ای دو رشته‌ای است. مهارکننده‌های RT غیرنوکلئوزیدی نقش مهمی در درمان HIV دارند و مسئول مهار RT با مسدود کردن سایت آلوستریک آن هستند که خواص فیزیکوشیمیایی مانند هیدروفوبیکیت بالا و تعاملات کلیدی آمینواسید با پیوند هیدروژن را نشان می‌دهند. به همین دلیل، طراحی مولکول‌های جدید با حلقه‌های هتروسیکلیک نوآورانه که با این ویژگی‌ها سازگار هستند، می‌تواند مهارکننده‌های بالقوه جدیدی فراهم کند. برای مثال، حلقه‌های پنتاسیکلیک مانند تریازول و مشتقات آن مانند تریازولیدین‌دایون، که خواص فیزیکوشیمیایی دارند که می‌توانند با سایت آلوستریک RT تعامل کنند، می‌توانند یک دیدگاه جدید در طراحی مهارکننده‌های جدید RT غیرنوکلئوزیدی باشند. این کار ادبیات علمی مربوط به این ترکیبات را مرور می‌کند و می‌تواند به طراحی داروهای ضدویروس جدید کمک کند.
