چکیده اصلی

BACKGROUND: Mammary tumours are the most common neoplasm in female dogs. Canine mammary tumours (CMTs) are appropriate models for human studies. OBJECTIVES: The present study aimed to diagnose CMTs pathologically and compare different tumour types by immunohistochemistry (IHC). METHODS: Histological features of various tumours were recorded. Moreover, haematological, biochemical and immunohistochemical changes were evaluated in affected animals. RESULTS: Among 130 mastectomized dogs in veterinary hospitals in Tehran, Iran, 74 dogs were affected by various types of mammary tumours. The highest frequency of malignant tumours was related to carcinoma types (38/74; 51.4%) and the highest frequency of benign tumours was related to fibroadenoma (6/74; 8.1%). In malignant tumours, anaemia and monocytosis were observed compared to benign tumours. Serum ALT and ALP levels were also significantly increased in malignant tumours (p < 0.05) compared to benign tumours. The IHC showed the profile of adenoma (ductal and intraductal papillary) ER+/PR-/HER2-, benign mixed tumour ER-/PR-/HER2-, carcinoma (mucinous, anaplastic) ER-/PR-/HER2- and metastatic mast cell tumour ER-/PR+/HER2-. The highest expression of αSMA was recorded in benign tumours and the highest expression of Ki67 was recorded in metastatic mast cell tumour, benign mixed tumour and adenoma (p < 0.05). CONCLUSION: Together, although the dog is a good model for studying mammary tumours, the variability of results in CMTs requires further studies in this field. An interesting finding of this study was the triple-negative profile in the benign mixed tumour, which had previously been reported only for malignant mammary tumours. In addition, the significant positivity of Ki67 expression (34%-66%) in benign CMTs is noted for the first time.

تومورهای پستان در سگ‌های ماده شایع‌ترین نئوپلاسم هستند. این مطالعه ۱۳۰ سگ subjected به mastectomy را بررسی کرد و ۷۴ مورد تومور پستان را شناسایی نمود. تومورهای بدخیم بیشتر کارسینوماتیک و تومورهای خوش‌خیم بیشتر فیبرآدنوما بودند. تومورهای بدخیم با آنمی، مونوسیتوز و افزایش ALT/ALP همراه بودند. یافته‌ای جالب، وجود پروفایل سه منفی (ER-/PR-/HER2-) در تومورهای خوش‌خیم بود که قبلاً فقط برای تومورهای بدخیم گزارش می‌شد.

تومورهای پستان شایع‌ترین نئوپلاسم در سگ‌های ماده هستند. این مطالعه با هدف تشخیص پاتولوژیک تومورهای پستان سگ (CMT) و مقایسه انواع مختلف تومور با استفاده از ایمونوهیستوشیمی (IHC) انجام شد. در میان ۱۳۰ سگ subjected به mastectomy در بیمارستان‌های دامی تهران، ۷۴ سگ به انواع مختلف تومور پستان مبتلا بودند. بیشترین فراوانی تومورهای بدخیم به انواع کارسینوماتیک مربوط بود (۳۸/۷۴؛ ۵۱.۴٪) و بیشترین فراوانی تومورهای خوش‌خیم به فیبرآدنوما (۶/۷۴؛ ۸.۱٪) تعلق داشت. در تومورهای بدخیم، آنمی و مونوسیتوز نسبت به تومورهای خوش‌خیم مشاهده شد. همچنین سطوح ALT و ALP در سرم در تومورهای بدخیم به طور معنی‌داری افزایش یافت (p < 0.05). IHC نشان داد که پروفایل adenoma (ductal و intraductal papillary) ER+/PR-/HER2-، benign mixed tumour ER-/PR-/HER2-، carcinoma (mucinous و anaplastic) ER-/PR-/HER2- و metastatic mast cell tumour ER-/PR+/HER2- است. بالاترین بیان αSMA در تومورهای خوش‌خیم و بالاترین بیان Ki67 در metastatic mast cell tumour، benign mixed tumour و adenoma ثبت شد (p < 0.05). نتیجه‌گیری: اگرچه سگ مدل خوبی برای مطالعه تومورهای پستان است، اما تنوع نتایج در CMTs نیاز به مطالعات بیشتر در این زمینه دارد. یک یافته جالب این مطالعه، پروفایل سه منفی در benign mixed tumour بود که قبلاً فقط برای تومورهای پستان بدخیم گزارش شده بود. علاوه بر این، مثبت بودن معنی‌دار بیان Ki67 (۳۴%-۶۶٪) در CMTs خوش‌خیم برای اولین بار ذکر شده است.

روش پژوهش

ویژگی‌های هیستولوژیک انواع تومورها ثبت شد. تغییرات هماتولوژیک، بیوشیمیایی و ایمونوهیستوشیمی در حیوانات مبتلا ارزیابی شد.

محدودیت‌ها

تنوع نتایج در CMTs نیاز به مطالعات بیشتر در این زمینه دارد. داده‌ها صرفاً از بیمارستان‌های دامی تهران جمع‌آوری شده‌اند.
