چکیده اصلی

INTRODUCTION: Metabolic stress increases Alzheimer's disease (AD) risk, but mechanisms linking metabolic dysfunction to neuroimmune dysregulation remain unclear. METHODS: We assessed cognition in community-dwelling participants and used high-fat diet (HFD)-fed 5xFAD mice, high glucose and high palmitate (HGHP)-treated regulatory T cell (Treg) co-cultures, and adoptive Treg transfer to evaluate metabolic stress-associated neuroimmune dysfunction. Flow cytometry, RNA sequencing, positron emission tomography computed tomography, and electrophysiology assessed Treg function, AD pathology, cerebral metabolism, and synaptic plasticity. RESULTS: Metabolic stress impaired Treg stability and function, accompanied by increased Z-DNA-binding protein 1 (ZBP1) expression and activation of apoptotic and necroptotic signaling. Treg dysfunction was associated with enhanced glial activation and neuronal injury. Under HGHP stress, ZBP1-deficient Tregs showed improved survival and stronger immunosuppressive activity, more effectively limiting glial activation and preserving neuronal integrity. In vivo, ZBP1-deficient Treg transfer conferred greater neuroprotection than wild-type Treg transfer in HFD-fed AD mice. DISCUSSION: ZBP1 may contribute to metabolic stress-induced Treg dysfunction and represent a candidate target to improve Treg-based therapy in HFD-fed AD mice.

مطالعه‌ای برای بررسی مکانیسم‌های پیوند اختلال متابولیک با اختلال سیستم عصبی-ایمنی در بیماری آلزایمر انجام شد. در این مطالعه، عملکرد شناختی در شرکت‌کنندگان جامعه‌نشین ارزیابی شد و از موش‌های 5xFAD تغذیه شده با رژیم غذایی چرب بالا (HFD)، هم‌کشت‌های سلولی Treg درمان شده با گلوکز و پالمیتیک اسید بالا (HGHP) و انتقال آداپتیو Treg استفاده شد. نتایج نشان داد که استرس متابولیک عملکرد و پایداری Treg را مختل کرده و بیان و فعال‌سازی ZBP1 را افزایش داده است. ناتوانی Treg با فعال‌سازی بیشتر گلیا و آسیب عصبی همراه بود. در شرایط استرس HGHP، Treg‌های فاقد ZBP1 مقاومت بیشتری داشتند و فعالیت سرکوب‌کننده ایمنی قوی‌تری از خود نشان دادند. انتقال Treg فاقد ZBP1 در موش‌های آلزایمر تغذیه شده با HFD، محافظت عصبی بیشتری نسبت به انتقال Treg وحشی‌نژاد ایجاد کرد.

روش پژوهش

ارزیابی عملکرد شناختی در شرکت‌کنندگان جامعه‌نشین و استفاده از موش‌های 5xFAD تغذیه شده با رژیم غذایی چرب بالا، هم‌کشت‌های سلولی Treg و انتقال آداپتیو Treg. استفاده از روش‌های سیتومتری جریان، توالی‌یابی RNA، PET-CT و الکتروفیزیولوژی.

محدودیت‌ها

مطالعه در موش‌های مدل انجام شده است و نتایج ممکن است در انسان تکرار نشود. مکانیسم دقیق نقش ZBP1 در این فرآیند هنوز کاملاً روشن نیست.
