چکیده اصلی

Auricularia polytricha is an economically important edible and medicinal mushroom widely cultivated in China. Ear thickness, as a core commercial trait of this mushroom, directly determines its product quality and economic value. To explore the potential molecular regulatory mechanisms governing ear thickness, we performed comparative transcriptome sequencing on fruiting bodies of the main cultivar 'Zhang'er 4328' and its stable thick-ear mutant 'B2872' at three key developmental stages: primordium stage (PD), young ear stage (IF), and mature stage (MF). Differentially expressed genes (DEGs) were screened, followed by Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses. GO enrichment results showed that DEGs were mainly enriched in biological processes associated with metabolic activity, cellular component formation, and catalytic function. KEGG enrichment analysis revealed a distinct stage-specific pattern: DEGs at the PD stage were primarily enriched in energy and substance synthesis pathways (e.g., thiamine metabolism, carbon metabolism); at the IF stage, DEGs were concentrated in pathways related to cell proliferation and structure formation (e.g., amino acid metabolism, nitrogen metabolism); and at the MF stage, DEGs were enriched in pathways involved in protein synthesis and cellular homeostasis maintenance (e.g., ribosome, endoplasmic reticulum protein processing). Our findings indicate that pathways related to energy metabolism, cell proliferation, and protein synthesis provide essential energy and material support for ear thickness in A. polytricha. This exploratory study preliminarily fills the research gap in the molecular mechanism underlying ear thickness variation in A. polytricha and provides a preliminary theoretical reference for molecular marker-assisted breeding of thick-ear varieties.

Auricularia polytricha یک قارچ خوراکی و دارویی مهم اقتصادی است که در چین کشت می‌شود. ضخامت گوش، یک ویژگی تجاری اصلی این قارچ، مستقیماً کیفیت محصول و ارزش اقتصادی آن را تعیین می‌کند. برای بررسی مکانیسم‌های تنظیمی مولکولی بالقوه governing ضخامت گوش، ترنسکریپتوم‌سنجی مقایسه‌ای بر روی بدنه‌های میوه‌دهی cultivar اصلی «Zhang'er 4328» و mutant ضخیم‌گوش پایدار «B2872» در سه مرحله کلیدی رشدی: مرحله بذر (PD)، مرحله گوش جوان (IF) و مرحله بلوغ (MF) انجام شد. ژن‌های بیان‌شده متفاوت (DEGs) غربالگری شدند و تحلیل‌های غنی‌سازی GO و KEGG انجام گرفت. نتایج GO نشان داد که DEGs عمدتاً در فرآیندهای زیستی مرتبط با فعالیت متابولیک، تشکیل مولکولات سلولی و عملکرد کاتالیتیک غنی شدند. تحلیل KEGG الگوی مرحله‌به‌مرحله متمایزی را آشکار کرد: DEGs در مرحله PD عمدتاً در مسیرهای سنتز انرژی و مواد غنی شدند (مثل متابولیسم تیامین و متابولیسم کربن)؛ در مرحله IF، DEGs در مسیرهای مرتبط با تکثیر سلولی و تشکیل ساختار متمرکز بودند (مثل متابولیسم اسیدهای آمینه و متابولیسم نیتروژن)؛ و در مرحله MF، DEGs در مسیرهای درگیر در سنتز پروتئین و حفظ هموستازی سلولی غنی شدند (مثل ریبوزوم و پردازش پروتئین رتیکولوم). یافته‌های ما نشان می‌دهد که مسیرهای مرتبط با متابولیسم انرژی، تکثیر سلولی و سنتز پروتئین پشتیبانی انرژی و متریال ضروری برای ضخامت گوش در A. polytricha فراهم می‌کنند. این مطالعه اکتشافی شکاف تحقیقاتی در مکانیسم مولکولی پشت تغییرات ضخامت گوش را در A. polytricha تا حدی پر می‌کند و یک مرجع نظری اولیه برای breeding ژنوم‌یارک کمک‌دار ضخامت‌گوش فراهم می‌کند.

روش پژوهش

ترنسکریپتوم‌سنجی مقایسه‌ای بر روی بدنه‌های میوه‌دهی cultivar اصلی و mutant ضخیم‌گوش در سه مرحله رشدی انجام شد. ژن‌های بیان‌شده متفاوت (DEGs) غربالگری شدند و تحلیل‌های غنی‌سازی GO و KEGG برای شناسایی مسیرهای مرتبط انجام گرفت.
