چکیده اصلی

CONTEXT: Mitragyna speciosa (Korth.) commonly known as 'kratom' is a tropical tree native to southeast Asia, that produces psychoactive monoterpenoid indole alkaloids including mitragynine. Kratom products are growing in popularity for increasing energy, improving mood and alleviating minor pain, but availability of commercial plant material is limited in North America. Propagation relies primarily on shoot cuttings rooted in soils with high levels of humus and is relatively inefficient. OBJECTIVE: This study aimed to optimize in vitro shoot induction system for kratom using a Design of Experiments (DOE) approach and compare the metabolite profiles of in vitro and greenhouse-grown plants using metabolomics. MATERIALS AND METHODS: A DOE approach was used to minimize the number of treatments necessary to optimize in vitro culture media to induce shoot and root morphogenesis in kratom explants. A clean mother stock of in vitro cultures was established and evaluated for medicinal bioactives via targeted and untargeted analyses by ultra-performance liquid chromatography tandem mass spectrometry (UPLC-MS/MS). RESULTS: We found no significant difference in the quantified levels of mitragynine, melatonin, and some related metabolites, between in vitro-grown and greenhouse-grown plants within the experimental conditions of this study. DISCUSSION AND CONCLUSIONS: The presence of similar specialized metabolite profiles in greenhouse and in vitro-grown kratom plants paves the way for further studies on biotechnological engineering and production of these valuable compounds. In vitro propagation is a viable approach to produce bioactive compounds in kratom plants.

این مطالعه از روش طراحی آزمایش (DOE) برای بهینه‌سازی کشت درون‌کشتگاهی گیاهان کرامات استفاده کرد. نتایج نشان داد که سطح میتراگینین و متابولیت‌های دیگر در گیاهان درون‌کشتگاهی و گلخانه‌ای تفاوتی معنی‌دار نداشت. این یافته‌ها نشان می‌دهد که تکثیر درون‌کشتگاهی می‌تواند برای تولید ترکیبات دارویی کارآمد باشد.

Mitragyna speciosa (Korth.) که به 'kratom' معروف است، درخت گرمسیری بومی جنوب شرق آسیا است که آلکالوئیدهای تریپتامین‌مانند موهومونوترپن‌های روان‌گردان، از جمله میتراگینین، تولید می‌کند. محصولات کرامات برای افزایش انرژی، بهبود خلق و خو و تسکین درد جزئی محبوبیت در حال افزایش دارند، اما در دسترس بودن مواد گیاهی تجاری در آمریکای شمالی محدود است. تکثیر عمدتاً به برش‌های شاخه‌ای ریشه‌دار در خاک‌های حاوی ساژ وابسته است و نسبتاً ناکارآمد است. هدف این مطالعه بهینه‌سازی سیستم القای شاخه‌های درون‌کشتگاهی برای کرامات با استفاده از رویکرد طراحی آزمایش (DOE) و مقایسه پروفایل متابولیتیک گیاهان درون‌کشتگاهی و گلخانه‌ای با استفاده از متابولومیک بود. یک رویکرد DOE برای کاهش تعداد درمان‌های لازم برای بهینه‌سازی رسانه‌های کشت درون‌کشتگاهی برای القای مورفوژنز شاخه و ریشه در نمونه‌های کرامات استفاده شد. یک مادر استریل از کشت‌های درون‌کشتگاهی تأسیس و برای بیواکتیوهای دارویی با تحلیل‌های هدفمند و غیرهدفمند با استفاده از اسپکترومتروگرافی جرمی مایع فوق‌سریع (UPLC-MS/MS) ارزیابی شد. ما هیچ تفاوت معنی‌داری در سطوح کمی‌سازی شده میتراگینین، ملاتونین و برخی متابولیت‌های مرتبط بین گیاهان درون‌کشتگاهی و گلخانه‌ای در شرایط آزمایشی این مطالعه پیدا نکردیم. حضور پروفایل‌های متابولیتیک تخصصی مشابه در گیاهان کرامات گلخانه‌ای و درون‌کشتگاهی، راه را برای مطالعات بیشتر در مهندسی بیوتکنولوژیک و تولید این ترکیبات ارزشمند هموار می‌کند. تکثیر درون‌کشتگاهی یک رویکرد پذیرفتنی برای تولید ترکیبات بیواکتیو در گیاهان کرامات است.

روش پژوهش

از روش طراحی آزمایش (DOE) برای بهینه‌سازی رسانه‌های کشت درون‌کشتگاهی استفاده شد. تحلیل‌های هدفمند و غیرهدفمند با اسپکترومتروگرافی جرمی مایع فوق‌سریع (UPLC-MS/MS) برای ارزیابی مادر استریل و بیواکتیوهای دارویی انجام شد.
