چکیده اصلی

This study investigated the synergistic antibacterial mechanism of ginger essential oil (GEO) and rosemary essential oil (REO) against Shewanella putrefaciens. Antibacterial efficacy and interactions were assessed by determining the minimum inhibitory concentration (MIC), fractional inhibitory concentration (FIC) index, and growth curves. Membrane integrity was evaluated using confocal laser scanning microscopy (CLSM), leakage of intracellular components, and alkaline phosphatase (AKPase) activity. Structural damage was observed via scanning electron microscopy (SEM). The effects on antioxidant defense and energy metabolism were analyzed by measuring superoxide dismutase (SOD), succinate dehydrogenase (SDH), and malate dehydrogenase (MDH) activities. Biofilm inhibition capacity was also examined. Results demonstrated a synergistic effect (FIC index = 0.375). The combination of GEO and REO caused severe membrane damage, intracellular leakage, inhibition of key enzyme activities, and effective biofilm inhibition. The synergistic mechanism involves membrane disruption, suppression of metabolic pathways, and biofilm inhibition.

این مطالعه نشان داد که ترکیب روغن‌های اسانسی گینجر و رزماری علیه Shewanella putrefaciens دارای اثر هم‌افزا است. این ترکیب باعث آسیب به غشای سلولی، نشت مواد داخل سلولی و مهار آنزیم‌های کلیدی می‌شود. همچنین، ظرفیت مهار بیوفیلم این ترکیب تأیید شده است.

این مطالعه مکانیسم آنتی‌باکتریال هم‌افزا روغن‌های اسانسی گینجر (GEO) و رزماری (REO) علیه Shewanella putrefaciens را بررسی کرد. اثربخشی آنتی‌باکتریال و تعاملات با تعیین غلظت مهارکننده حداقل (MIC)، شاخص غلظت مهارکننده کسری (FIC) و نمودارهای رشد ارزیابی شد. یکپارچگی غشا با میکروسکوپ سکانر لیزر کانفوکال (CLSM)، نشت اجزای سلولی و فعالیت آنزیم فسفاتاز قلیایی (AKPase) ارزیابی شد. آسیب ساختاری با میکروسکوپ الکترونی اسکن (SEM) مشاهده شد. اثرات بر دفاع آنتی‌اکسیدانی و متابولیسم انرژی با اندازه‌گیری فعالیت‌های سوپراکسید دی‌موتاز (SOD)، دی‌هیدروژناز سیترات (SDH) و دی‌هیدروژناز مالات (MDH) تحلیل شد. ظرفیت مهار بیوفیلم نیز بررسی شد. نتایج نشان‌دهنده اثر هم‌افزا (شاخص FIC = 0.375) بود. ترکیب GEO و REO باعث آسیب شدید غشا، نشت داخل سلولی، مهار فعالیت آنزیم‌های کلیدی و مهار مؤثر بیوفیلم شد. مکانیسم هم‌افزا شامل اختلال غشا، مهار مسیرهای متابولیکی و مهار بیوفیلم است.

روش پژوهش

اثربخشی آنتی‌باکتریال با تعیین MIC و FIC و نمودارهای رشد ارزیابی شد. آسیب ساختاری با SEM و میکروسکوپ کانفوکال بررسی شد. فعالیت آنزیم‌های SOD، SDH و MDH برای ارزیابی اثرات بر دفاع آنتی‌اکسیدانی و متابولیسم انرژی اندازه‌گیری شد.
