چکیده اصلی

To address the challenge of fungicide resistance in plant pathogenic fungi, coumarin derivatives were designed. Two series of coumarin derivatives, including 3-amide and thienocoumarin ether derivatives, were designed and synthesized through structural modification of the coumarin scaffold to explore their antifungal potential. Their antifungal activities were evaluated using the mycelial growth rate method, with half-maximal effective concentration (EC50) values determined. The effects of compound IVd on membrane integrity and morphology were assessed by measuring electrolyte leakage, malondialdehyde (MDA) content, and scanning electron microscopy (SEM). Synergistic effects were quantified using Wadley's method by calculating the synergistic ratio (SR). Molecular docking studies were conducted to investigate interactions with potential targets (CYP51, CYP51B, and SDH). Among the derivatives, IVd showed the most potent antifungal activity, with EC50 values of 88.5 µg/mL against Valsa mali, slightly surpassing azoxystrobin (EC50 = 92.8 µg/mL). Treatment with IVd caused membrane disruption and hyphal deformation. A 2:1 combination ratio of IVd with azoxystrobin or fluconazole demonstrated synergistic effects (SR > 1.5). Molecular docking suggested favorable interactions with the investigated targets, supporting its multitarget activity. Overall, these findings highlight IVd as a promising antifungal scaffold for investigation and provide evidence for its potential use in combination strategies against plant pathogenic fungi.

مشتق کومارین IVd با غلظت مؤثر نیمی (EC50) ۸۸.۵ میکروگرم بر میلی‌لیتر در برابر Valsa mali، فعالیت ضدقارچی قوی‌تری نسبت به آزوکستروبین نشان داد. این ماده باعث تخریب غشا و تغییر شکل هیف می‌شود. ترکیب IVd با آزوکستروبین یا فلوکونازول در نسبت ۲:۱ اثرات هم‌افزا ایجاد می‌کند. مطالعات بندل‌سازی نشان‌دهنده تعامل مطلوب با اهداف CYP51 و SDH است.

برای مواجهه با چالش مقاومت قارچ‌های بیماری‌زا گیاهی، مشتقات کومارین طراحی شدند. دو سری مشتق کومارین، شامل مشتقات آمید ۳ و اتر تیینوکومارین، از طریق اصلاح ساختاری اسکلت کومارین طراحی و سنتز شدند تا پتانسیل ضدقارچی آن‌ها بررسی شود. فعالیت ضدقارچی آن‌ها با استفاده از روش رشد هیف بررسی شد و مقادیر غلظت مؤثر نیمی (EC50) تعیین گردید. اثرات مشتق IVd بر پایداری و ظاهر غشا با اندازه‌گیری نشت الکترولیت، محتوای مالون‌دی‌آلدهید (MDA) و میکروسکوپ الکترونی اسکن (SEM) ارزیابی شد. اثرات هم‌افزا با استفاده از روش Wadley و محاسبه نسبت هم‌افزا (SR) کمی‌سازی شدند. مطالعات بندل‌سازی مولکولی برای بررسی تعامل با اهداف بالقوه (CYP51، CYP51B و SDH) انجام شد. در میان مشتقات، IVd قوی‌ترین فعالیت ضدقارچی را نشان داد، با مقادیر EC50 برابر با ۸۸.۵ میکروگرم بر میلی‌لیتر در برابر Valsa mali، که کمی از آزوکستروبین (EC50 = ۹۲.۸ میکروگرم بر میلی‌لیتر) فراتر رفت. درمان با IVd باعث تخریب غشا و تغییر شکل هیف شد. نسبت ترکیبی ۲:۱ از IVd با آزوکستروبین یا فلوکونازول اثرات هم‌افزا را نشان داد (SR > ۱.۵). بندل‌سازی مولکولی تعاملات مطلوبی با اهداف بررسی شده را نشان داد که فعالیت چندهدفه آن را پشتیبانی می‌کند. در مجموع، این یافته‌ها IVd را به عنوان یک اسکلت ضدقارچی امیدوارکننده برای بررسی و شواهدی برای استفاده بالقوه در استراتژی‌های ترکیبی در برابر قارچ‌های بیماری‌زا گیاهی ارائه می‌دهند.
