چکیده اصلی

This study evaluated the antibacterial effect of photodynamic therapy (aPDT) using nanomicelle-encapsulated curcumin photosensitizer (PS) against drug-resistant clinical isolates of Klebsiella pneumoniae and K. pneumoniae ATCC 13883™. In this study, nanomicelle curcumin prepared by the thin film hydration method, with LED as the radiation source, was used for effective aPDT. The technique's effectiveness was analysed through membrane changes tracked by flow cytometry, bacterial structural alterations observed via FTIR, lipid peroxidation quantification assay, atomic force microscopy, and its performance against 17 clinical isolates resistant to cephalosporins and carbapenems was evaluated. Based on the results of aPDT, the minimum inhibitory concentration of the PS was 200 μg/mL for the standard strain and 125-1000 μg/mL for superbug clinical isolates. With an irradiation dose of 500 J/cm2, an irradiation time of 500 s, and a power density of 1000 mW/cm2 from an LED, bacterial growth was inhibited by a mean of 8 Log10 CFU/mL reduction, as determined by the culture method. Additionally, the results of the bacterial structural integrity assays (flow cytometry, FTIR, MDA levels, and AFM) for the standard strain provided convergent evidence with the culture results. This study demonstrates antibacterial activity of LED-illuminated curcumin nanomicelles against multidrug-resistant K. pneumoniae.

این مطالعه اثر درمان فتو دینامیک (aPDT) با نانومیسل‌های کورکومین روشن شده با LED را در برابر کلامیلا پنومونیای مقاوم به دارو بررسی کرد. نتایج نشان داد که این روش باکتری‌ها را با کاهش ۸ Log10 CFU/میلی‌لیتر مهار می‌کند. آزمایش‌های مولکولی و میکروسکوپی نیز تغییرات ساختاری و غشایی باکتری را تأیید کردند.

این مطالعه اثر آنتی‌باکتریال درمان فتو دینامیک (aPDT) با استفاده از حساس‌ساز فتون کورکومین محصور شده در نانومیسل در برابر ایزولت‌های بالینی مقاوم به دارو و کلامیلا پنومونیای ATCC 13883 ارزیابی کرد. نانومیسل کورکومین آماده شده با روش هیدراتاسیون فیلم نازک و LED به عنوان منبع تابش برای aPDT مؤثر استفاده شد. اثربخشی تکنیک از طریق تغییرات غشای ردیابی شده با سیتومتری جریان، تغییرات ساختاری باکتریایی مشاهده شده با FTIR، اندازه‌گیری آنزیم پراکسیداسیون لیپید، میکروسکوپ نیرو اتمی و عملکرد آن در برابر ۱۷ ایزولت بالینی مقاوم به سفتاپنوم‌ها و کرباپنمز ارزیابی شد. بر اساس نتایج aPDT، غلظت مهارکننده حداقل (MIC) حساس‌ساز فتون ۲۰۰ میکروگرم/میلی‌لیتر برای استاندارد و ۱۲۵-۱۰۰۰ میکروگرم/میلی‌لیتر برای ایزولت‌های بالینی سوپربیوکتریال بود. با دوز تابش ۵۰۰ جول/سانتی‌متر مربع، زمان تابش ۵۰۰ ثانیه و چگالی توان ۱۰۰۰ میلی‌وات/سانتی‌متر مربع از LED، رشد باکتری با میانگین کاهش ۸ Log10 CFU/میلی‌لیتر مهار شد. همچنین نتایج آزمون‌های پایداری ساختاری باکتریایی (سیتومتری جریان، FTIR، سطح MDA و AFM) برای استاندارد شواهدی همگرا با نتایج کشت ارائه داد. این مطالعه فعالیت آنتی‌باکتریال نانومیسل‌های کورکومین روشن شده با LED در برابر کلامیلا پنومونیای چنددارویی مقاوم را نشان می‌دهد.

روش پژوهش

نانومیسل کورکومین با روش هیدراتاسیون فیلم نازک و LED به عنوان منبع تابش آماده شد. اثربخشی با روش‌های سیتومتری جریان، FTIR، آنزیم پراکسیداسیون لیپید، میکروسکوپ نیرو اتمی و کشت ارزیابی شد.
