Once considered somewhere between inert ('junk DNA') and parasitic ('selfish genetic elements'), transposable elements (TEs) are increasingly appreciated as inseparable from the healthy functioning of eukaryotic genomes. The ubiquitous TE sequences that make up about half of the human genome are sources of the physical genetic material necessary for establishing distinct domains of chromatin architecture, generating non-coding RNAs (ncRNAs), expanding transcription factor regulatory networks, and providing cis-regulatory elements to enable complex gene expression patterns. Given the critical roles for TEs in core genomic functions, the circumstances in which TEs become dysregulated are increasingly being explored in the context of human disease. While most TEs are silenced by host-mediated epigenetic and transcriptional mechanisms, certain tissues and cell types are noted to experience considerable TE transcription. The brain and nervous tissues notably exhibit highly dynamic TE activity, both in response to experience and across the lifespan. Here, we review the current state of research into epigenetic regulation of and by TEs with a focus on the brain. We explore the intersection of epigenetics and TEs in brain cell types, with emphasis on DNA and histone methylation, ncRNAs and small RNAs, and the function of TE-regulating transcription factors. We explore how epigenetic mechanisms involved in brain TE regulation may become dysfunctional in the context of neurodevelopmental, neurodegenerative, and neuropsychiatric disorders. We present and synthesize the mounting evidence implicating the epigenetic control of brain TEs as an under-explored mechanism in these complex disorders.

AA and Parkinson's Disease.

Parkinson's disease (PD) is due to degeneration of dopaminergic neurons in substantia nigra pars compacta. PD neuropathology is characterized by the presence of Lewy bodies that contain aggregated α-synuclein, which has been implicated in a range of interactions with phospholipid membranes and free fatty acids. I propose that α-synuclein interacts with arachidonic acid (AA) to regulate the function of dopaminergic neurons. α-Synuclein …

AA in Brain-Memory, Dementia, Depression, Anxiety, Alzheimer's Disease, Affective and Psychiatric Disorders, and Coronary Heart Disease (CHD).

The human brain is rich in AA and DHA, which are crucial for normal brain growth and development and memory. They form a major structural component of neurons of the cerebral cortex and photoreceptor cells in the retina. AA and DHA support synaptic plasticity and contribute to efficient neurotransmission. Adequate amounts of AA and DHA support neuronal growth and the formation of new synapses and thus are crucial for memory and cogniti…

Systematic Dissection of Key Driver Perturbation Signatures in Single Cells via ECCITE-seq.

CRISPR screens, such as expanded CRISPR-compatible cellular indexing of transcriptomes and epitopes by sequencing (ECCITE-seq), enable the simultaneous measurement of transcriptomes, gRNA identity, and cell-surface protein expression at single-cell resolution to systematically interrogate gene function. This platform provides a powerful and scalable experimental approach for validating disease-associated regulators identified by large-…

'Got to be the right fit, the right connection': a scoping review of the peer support experiences and perspectives of adults with acquired neurological disability.

BACKGROUND: Peer support for people with acquired neurological disability is gaining worldwide popularity, yet it is conceptually unclear what constitutes valuable peer interactions. To build a greater understanding from the lived experience perspective, this scoping review synthesised literature on how adults with acquired neurological disability experience and perceive peer support. METHODS: Peer-reviewed literature published between…
