چکیده اصلی

INTRODUCTION: Polyphenols have been investigated in dental materials and restorative techniques due to their interactions with metal cations, antioxidant activity, and collagen crosslinking ability, which make them a promise for reinforcing the dental substrate and improving material adhesion. However, more studies are necessary before to support their use in clinical procedures. OBJECTIVES: This study evaluated the effects of quercetin and resveratrol on bond strength and the protection of dentin collagen fibrils against enzymatic degradation. MATERIALS AND METHODS: Demineralized human dentin specimens were immersed for 1 min in solutions of quercetin or resveratrol (100, 250, and 500 µg/mL), 2% chlorhexidine (CHX), or 0.9% saline (control). The specimens were then incubated in collagenase for 24 h at 37°C, and changes in dry mass were measured. For microtensile bond strength testing, quercetin, resveratrol, CHX, or distilled water (DW) were applied to phosphoric acid-conditioned dentin, followed by application of Single Bond 2 (3 M/ESPE) adhesive and Z350XT resin (3 M/ESPE). Adhesive strength was measured after 24 h and after 1 year in distilled water. Data were analyzed using the Kruskal-Wallis and Dunn tests or ANOVA followed by Tukey's test (p < 0.05). RESULTS: Quercetin-treated specimens showed a significant weight increase compared with CHX (p < 0.05). Both quercetin- and resveratrol-treated groups exhibited resistance to collagenase similar to CHX (p > 0.05). After 1 year, only the DW group showed a significant decrease in bond strength (p = 0.001). CONCLUSIONS: Dentin treatment with quercetin or resveratrol effectively preserved adhesive strength and protected collagen against enzymatic degradation, suggesting their potential as agents to improve the longevity of adhesive restorations.

این مطالعه اثر کئرستین و رزوراترول بر استحکام پیوند و محافظت کلاژن دندان‌سوخت را بررسی کرد. نتایج نشان داد که هر دو ماده در حفظ استحکام پیوند و مقاومت در برابر آنزیم کلاژناز موثر بودند. پس از یک سال، تنها گروه کنترل کاهش معنی‌داری در استحکام پیوند داشت.

مقدمه: پلی‌فنول‌ها به دلیل تعامل با یون‌های فلزی، فعالیت آنتی‌اکسیدانی و توانایی تقویت پیوندهای کلاژن، در مواد دندان‌پزشکی و تکنیک‌های ترمیمی مورد بررسی قرار گرفته‌اند. هدف: این مطالعه اثر کئرستین و رزوراترول بر استحکام پیوند و محافظت فیبرهای کلاژن دندان‌سوخت در برابر تخریب آنزیمی را ارزیابی کرد. روش‌ها: نمونه‌های دندان‌سوخت انسان دمینرالیزه به مدت ۱ دقیقه در محلول‌های کئرستین یا رزوراترول (۱۰۰، ۲۵۰ و ۵۰۰ میکروگرم/میلی‌لیتر)، ۲ درصد کلرهکسی‌دین (CHX) یا ۰.۹ درصد نمک غلیظ (کنترل) غوطه‌ور شدند. نمونه‌ها سپس در ۳۷ درجه سانتی‌گراد به مدت ۲۴ ساعت در آنزیم کلاژناز انکوبه شدند و تغییرات جرم خشک اندازه‌گیری شد. برای تست استحکام پیوند میکرومقاومتی، کئرستین، رزوراترول، CHX یا آب مقطر (DW) روی دندان‌سوخت آماده‌شده با اسید فسفریک اعمال شد، سپس چسب Single Bond 2 و رزین Z350XT اعمال گردید. استحکام چسبندگی پس از ۲۴ ساعت و یک سال در آب مقطر اندازه‌گیری شد. داده‌ها با آزمون‌های کروسکال-والیس و دان یا ANOVA و آزمون Tukey تحلیل شدند (p < 0.05). نتایج: نمونه‌های درمان‌شده با کئرستین افزایش معنی‌داری در جرم داشتند در مقایسه با CHX (p < 0.05). هر دو گروه درمان‌شده با کئرستین و رزوراترول مقاومت مشابهی در برابر آنزیم کلاژناز نشان دادند که مشابه CHX بود (p > 0.05). پس از یک سال، تنها گروه DW کاهش معنی‌داری در استحکام پیوند داشت (p = 0.001). نتیجه‌گیری: درمان دندان‌سوخت با کئرستین یا رزوراترول به طور مؤثر استحکام چسبندگی را حفظ کرد و کلاژن را در برابر تخریب آنزیمی محافظت نمود، که نشان‌دهنده پتانسیل آن‌ها به عنوان عوامل بهبود طول عمر ترمیم‌های چسبانی است.
