چکیده اصلی

Peripheral nerve defect repair remains a clinical challenge. Nerve guidance conduits (NGCs) are potential alternatives to autografts, but their performance can be compromised by the oxidative and inflammatory microenvironment after injury. Here, we fabricated MnO2-loaded hydrogel NGCs at concentrations of 10, 50, and 100 μg ml-1and evaluated them in a rat model of a 10-mm sciatic nerve defect. Among the three concentrations, the MnO2@NMgel-10 (10 μg ml-1) group yielded a higher sciatic functional index, denser and more orderly regenerated nerve tissue, and less gastrocnemius muscle atrophy than the hydrogel-only and higher-dose conduit groups. Early tissue analysis showed that the MnO2@NMgel-10 group exhibited increased infiltration of CD86+CD206+cells, upregulated expression of IL-10, and significantly downregulated expression of IL-1β, TNF-α, and 4-HNE. Furthermore, the MnO2@NMgel-10 group showed satisfactory biosafety and biocompatibility. These findings reveal a non-monotonic concentration response and preliminarily confirm the feasibility of low-dose MnO2-functionalized hydrogel NGCs for repairing peripheral nerve defects, providing new insights into the application of MnO2as a supplementary material for NGC fabrication.

بازسازی نقص عصب محیطی همچنان یک چالش بالینی باقی مانده است. مجاری هدایت عصب (NGC) به عنوان جایگزین‌های بالقوه برای graft خودبخودی عمل می‌کنند، اما عملکرد آن‌ها می‌تواند تحت تأثیر محیط اکسیداتیو و التهابی پس از آسیب قرار گیرد. در اینجا، ما مجاری NGC حاوی هیدروژل بارگذاری شده با MnO2 در غلظت‌های 10، 50 و 100 μg ml-1 ساختیم و آن‌ها را در مدل موش با نقص 10 میلی‌متری عصب ساق پا ارزیابی کردیم. در میان سه غلظت، گروه MnO2@NMgel-10 (10 μg ml-1) شاخص عملکردی عصب ساق پای بالاتر، بافت عصب بازسازی شده متراکم‌تر و منظم‌تر و آتروفی عضله گاستروکنمیوس کمتری نسبت به گروه‌های مجاری فقط هیدروژل و با دوز بالاتر ارائه داد. تحلیل بافت زودهنگام نشان داد که گروه MnO2@NMgel-10 نفوذ سلول‌های CD86+CD206+، بیان افزایش‌یافته IL-10 و کاهش معنی‌دار بیان IL-1β، TNF-α و 4-HNE را نشان داد. علاوه بر این، گروه MnO2@NMgel-10 خواص بیوسودار و بیولوژیک مناسب را نشان داد. این یافته‌ها یک پاسخ غلظتی غیرخطی را آشکار می‌کنند و اولیهً قابلیت استفاده از هیدروژل NGC تزئین شده با منگنز دی‌اکسید با دوز پایین را برای ترمیم نقص عصب محیطی تأیید می‌کنند.

روش پژوهش
